Brothers is presented from a third-person view overlooking the two brothers. The brothers are moved individually by two thumbsticks on the controller. The controller triggers also cause the respective brother to interact with the game world, such as talking to a non-player character or grabbing onto a ledge or object. The older brother is the stronger of the two and can pull levers or boost his younger brother to higher spaces, while the younger one can pass between narrow bars. The player progresses by manipulating the two brothers at the same time to complete various puzzles..
两位孩子为了拯救他们身患重病的父亲，逼迫踏上了寻找传说中的“生命之水”（Water of Life）的道路，他们必须在互相依靠，互相帮助的状况下才能求得生存，在一个孩子虚弱时，另一个孩子就必须坚强，一个孩子恐惧时，另一个孩子就必须勇敢，玩家将会以这对拯救父亲的兄弟视角来经历一段神奇坎坷的冒险之旅。
